Progress in the study of lotus genetic linkage map

The genetic linkage map is a powerful tool for studying plant genome structure and evolution, and is an important foundation for gene positioning, cloning and molecular marker-assisted breeding. However, as one of the earliest plants in angiosperms, and known as the "living fossil" lotus, its genomics research has obviously lagged behind. Therefore, the construction of the genetic linkage map of lotus is helpful for the rapid development of lotus genomics, and provides a basis for the isolation and cloning of important economic traits of lotus, which is conducive to further exploration and full use of the ornamental, edible and medicinal value of lotus.

The aquatic plant resources discipline group of the Wuhan Botanical Garden of the Chinese Academy of Sciences first scanned the ancient Chinese lotus genome sequence and found 86,089 SSR sites, of which 73,246 SSR sites could be used to develop SSR primers. Then, using the F1 population of the Asian lotus variety "Ancient Chinese Lotus" and the American yellow lotus variety "AL1" as materials, a part of the SSR molecular markers (500 pairs) developed based on the sequence information of the ancient Chinese lotus genome were used to construct "Chinese ancient lotus" and The genetic linkage map of "AL1". Due to the low heterozygosity of the genome of "Ancient Chinese Lotus", this map contains only 7 linkage groups with 47 markers and the map length is 365.67 cM. The "AL1" map contains 11 linkage groups with 177 molecular markers and the map length is 524.51 cM. This is the first report of the genetic linkage map of lotus, which laid a good foundation for lotus genomics research and accelerated the process of lotus important gene mapping, cloning and molecular marker-assisted breeding.
